Hexarelin Peptide (Examorelin)

Description

Hexarelin (Examorelin) is a synthetic growth hormone–releasing peptide (GHRP) composed of six amino acids. It is structurally distinct yet functionally related to ghrelin, a 28–amino acid peptide known to stimulate growth hormone (GH) release and appetite. Hexarelin acts primarily through growth hormone secretagogue receptors (GHSR-1a) located in the pituitary and hypothalamus, initiating intracellular signaling cascades that may promote GH secretion.

Mechanism of Action

  • Mimics ghrelin by binding to GHSR-1a receptors.
  • Stimulates GH release via G-protein–mediated pathways (e.g., protein kinase C activation).
  • May also influence appetite-regulating neuropeptides (NPY, AgRP) and reduce α-MSH, shifting balance toward increased hunger.
  • Can transiently desensitize receptors with repeated exposure.
  • May induce secretion of other pituitary hormones (ACTH, prolactin), though these effects are generally undesired in experimental contexts.

Chemical Properties

    • Molecular Formula: C47H58N12O6
    • Molecular Weight: 887.05 g/mol
    • Synonyms: Examorelin

Research Findings (Preclinical Models)

    • Growth Hormone Release: Hexarelin has been shown to elevate GH levels in adolescent and mature models, with variable effects in elderly groups.
    • Synergism with GHRH: Evidence suggests Hexarelin and GHRH act via distinct pathways, potentially producing additive GH stimulation when combined.
    • Cardiovascular Activity: Acute administration may enhance cardiac output, stroke volume, and left ventricular ejection fraction, while reducing apoptosis in ischemic heart tissue.
    • Body Composition: GH release following Hexarelin exposure appears inversely correlated with fat mass; sex differences were not significant.
    • Muscle Tissue Preservation: Preliminary studies indicate Hexarelin may reduce muscle mass and strength loss under catabolic conditions.
  • Plain-Language Benefits (Research Context Only)

  • Supports growth hormone release – may stimulate GH secretion in laboratory models.
  • Cardiac effects – may improve heart function and protect cardiac cells in experimental studies.
  • Muscle preservation – may reduce muscle loss under catabolic stress.
  • Appetite regulation – may increase hunger signals and alter feeding behavior.
  • Potential synergism – may complement GHRH activity for enhanced GH release.
